Abstract: A bio-printed therapeutic dermal construct (100) designed to accelerate skin regeneration and wound healing through the integration of autologous skin-derived exosomes (102) within a biodegradable hydrogel scaffold (101). The construct comprises a multi-layered architecture (103), including a dermal layer mimic (103a) with an interconnected porous structure to promote cell infiltration and angiogenesis, and an epidermal barrier layer (103b) that provides mechanical reinforcement and infection protection. Fabrication involves extrusion-based bioprinting of a bio-ink containing Gelatin Methacryloyl and Alginate, followed by sequential ionic and photo-crosslinking to preserve exosome bioactivity. The construct is customized using patient-specific wound mapping data to achieve anatomical conformity and sustained exosome-mediated biological signalling. The system provides a dual therapeutic effect structural support and regenerative bio signalling offering a personalized, biocompatible, and effective solution for the treatment of chronic and acute wounds, such as diabetic ulcers and pressure sores.
