Abstract: The invention discloses a topical roll-on pharmaceutical composition comprising lidocaine or a pharmaceutically acceptable salt thereof entrapped in cubosomal nanoparticles formed from glyceryl monooleate and poloxamer 407. The cubosomal dispersion is incorporated in a hydroxypropyl methylcellulose gel matrix containing menthol, oleic acid, methyl paraben and purified water, and is dispensed through a roller-ball applicator. In an exemplary 30 mL batch, the composition contains 50 mg lidocaine, 700-900 mg glyceryl monooleate, 100-300 mg poloxamer 407, 2.5% hydroxypropyl methylcellulose, 5% menthol, 2% oleic acid and 0.01% methyl paraben. The process comprises melting the lipid at 60-70°C, incorporating lidocaine, adding an aqueous poloxamer phase, homogenizing or probe-sonicating to form cubosomes, equilibrating, blending with the gel matrix, adjusting the pH, and filling into a roll-on container. The composition provides nanosized, homogeneous and conveniently applicable topical delivery.
