Abstract: TOLFENAMIC ACID-LOADED BINARY ETHOSOMAL THERMOGEL FORMULATION FOR TRANSDERMAL DELIVERY The present invention relates to a Tolfenamic Acid-loaded binary ethosomal thermogel formulation for transdermal delivery. The invention provides a nano-based vesicular drug delivery system comprising binary ethosomes incorporated within a thermogel matrix for enhanced permeation, sustained release, and prolonged retention over the skin surface. The binary ethosomes enhance aqueous solubility and transdermal permeation of Tolfenamic Acid, while the thermogel undergoes rapid sol-to-gel transformation at physiological temperature. The formulation exhibits improved anti-inflammatory activity, enhanced flux and apparent permeability, immediate gelation, prolonged residence time, and sustained drug release up to 24 hours. The invention further provides a simple and cost-effective preparation method involving thin-film hydration technique and cold method-based thermogel preparation. The formulation reduces gastrointestinal side effects associated with oral administration and improves patient compliance.
