Abstract: The present invention relates to thermostable and dissolvable polymeric microneedle based transdermal vaccine delivery system. The system comprises a polymeric microneedle array loaded with one or more antigens and one or more stabilizing agents. The vaccine formulation comprises diphtheria toxoid, tetanus toxoid, pertussis antigens, inactivated poliovirus antigens and Haemophilus influenzae type b antigen together with stabilizing agents selected from sucrose, trehalose, arginine, Tween 20 and Tween 80 and combinations thereof. The vaccine loaded polymeric microneedle arrays shows adequate mechanical strength for skin penetration and improved antigen stability at 2–8 °C, 25 °C, 40 °C storage conditions for a period of about 21 days. The proposed thermostable and dissolvable polymeric microneedle vaccine delivery system provides a convenient, stable, and effective alternative to conventional injection-based vaccination methods and reduces dependence on cold chain system.
