Abstract: Thermo-Responsive In-Situ Gelling Formulation for Fluconazole-Based Antifungal Treatment This invention describes a thermo-responsive in-situ gelling formulation for the localized delivery of fluconazole, targeting the treatment of vaginal candidiasis. This formulation utilizes thermoreversible polymers, specifically Poloxamer 407 and Poloxamer 188, which undergo a sol-to-gel transition at physiological temperatures, ensuring prolonged retention and controlled drug release at the site of infection. The formulation is optimized to maintain vaginal pH (4.0–5.5), bioadhesive properties, and pseudoplastic flow for ease of application and enhanced therapeutic efficacy. By bypassing formulationic absorption and hepatic first-pass metabolism, the formulation minimizes formulationic side effects while achieving sustained antifungal activity for at least 2.5 hours. Prepared using a cold method to preserve stability and homogeneity, the formulation is housed in sterile, tamper-proof containers with precision applicators for accurate delivery. Scalable for pharmaceutical production, this innovative formulation offers an effective, patient-friendly, and commercially viable solution for antifungal therapy.
