Abstract: A therapeutic formulation for cancer immunotherapy and process thereof is provided. The therapeutic formulation includes mRNA encoding an immunogenic fragment of the tumor-associated antigen (TAA), encapsulated in lipid nanoparticles (LNPs) using a droplet-based microfluidic device. The invention introduces a co- administration of mRNA with a CTLA4 inhibitor amplifies the immune response by further unleashing T cell activity, enhancing the formulation’s ability to combat resistant or recurrent cancer cells. The invention provides a highly specific immune response against oral cancer by delivering mRNA encoding the tumor-associated antigen (TAA) to antigen-presenting cells (APCs), ensuring that immune cells selectively target and destroy cancer cells while sparing healthy tissue. The therapeutic formulation minimizes systemic toxicity and collateral damage, reducing adverse effects like immunosuppression and tissue damage.
