Abstract: The present invention discloses an integrated dual-mode optical microscopy system and method for simultaneous acquisition and fusion of structural and functional imaging data of biological specimens. The system comprises an illumination subsystem including an LED array for Fourier Ptychographic Phase Microscopy (FPPM) and a fluorescence excitation source, an optical subsystem with dichroic mirrors, beam splitters, optical filters, and a shared optical path, an image acquisition module, a synchronization controller, and a computational subsystem. The computational subsystem utilizes deep learning-based multimodal fusion comprising modality-specific encoders and a cross-attention fusion framework for generating intrinsically co-registered structural-functional images in real time. The invention further provides virtual staining capability for generating fluorescence-like contrast from label-free FPPM phase data while reducing photobleaching and phototoxicity through adaptive illumination control. The invention enables high-resolution, real-time multimodal biomedical imaging suitable for digital pathology, live-cell imaging, cancer diagnostics, neuroscience, and drug discovery applications.
