Abstract: The present invention relates to a novel chalcone derivative (E)-1-(4-(3-(4-acetylphenoxy) propoxy) phenyl)-3-phenylprop-2-en-1-one (CR3), having molecular formula C26H24O4 and molecular weight 400.47 g/mol. The compound features a unique bis-aryl ether architecture wherein a chalcone moiety and an acetyl-functionalized aromatic ring are connected through a propyleneoxy ether bridge. CR3 is synthesized through a three-step route involving Claisen-Schmidt condensation, O-alkylation with 1-bromo-3-chloropropane, and Williamson ether synthesis. The compound satisfies Lipinski rule of five with zero violations, exhibits high gastrointestinal absorption, blood-brain barrier permeability, and a bioavailability score of 0.55. Antimicrobial evaluation by broth microdilution reveals MIC values of 2.364, 2.123, and 3.413 mg/ml against E. coli, B. subtilis, and C. albicans respectively. The invention provides a novel antimicrobial lead compound with favourable drug-likeness properties. [Figure 1]
