Abstract: Obesity linked to gut microbiota dysbiosis demands safe, effective natural interventions. The present invention discloses a synbiotic anti-obesity composition comprising a standardized low-molecular-weight polysaccharide fraction (IO-PLF, 3 to 8 kDa) obtained from Indian Himalayan Inonotus obliquus through enzyme- assisted extraction and tangential-flow ultrafiltration, combined with Lactobacillus plantarum MTCC 2621 and Bifidobacterium longum MTCC 3152. In HFD-fed mice, the synbiotic composition reduced body weight gain to 35 percent versus 78 percent in HFD controls, lowered serum triglycerides, total cholesterol, and liver transaminases, and selectively suppressed Faecalibaculum rodentium while enriching Lactobacillaceae and Bifidobacteriaceae. A 12-week human pilot study (n equals 60) demonstrated 3.6 kilogram weight reduction, 18.6 percent triglyceride reduction, and significant fecal microbiota shifts. The composition is formulated as functional beverages, effervescent sachets, and enteric-coated capsules with 12-month shelf stability, representing a translatable synbiotic strategy for obesity prevention.
