Abstract: Conventional oral antidiabetic therapies may cause poor patient compliance, gastrointestinal side effects, and fluctuations in plasma drug concentrations. The invention relates to a sustained-release transdermal patch containing a gymnemic acid-rich fraction obtained from Gymnema sylvestre leaves. The fraction is prepared by ethanolic Soxhlet extraction, alkaline dissolution, acid precipitation, filtration, and drying. The patch is formed by solvent casting with HPMC K100, PEG 400, and oleic acid. Placebo and medicated formulations were evaluated for appearance, folding endurance, uniformity, handling, thickness, weight variation, drug content, and percentage yield. Drug release was studied using an egg membrane in a Franz diffusion cell, and ex-vivo permeation was studied using goat skin. The optimized formulation showed in-vitro and ex-vivo permeation values of 60.83 µg/cm² and 59.58 µg/cm², respectively. The patch extract produced 9.74% alpha-amylase inhibition under the reported assay conditions. Compared with the marketed antidiabetic tablet, the Gymnema sylvestre transdermal patch showed prolonged drug release, indicating sustained therapeutic effects, reduced concentration fluctuations, and improved patient compliance.
