Abstract: The present invention relates to the field of nanotechnology and pharmaceutical delivery systems, specifically to a surface-engineered polymeric nanocarrier designed for targeted brain drug delivery. The invention comprises a biocompatible and biodegradable polymeric nanocarrier with an amphiphilic structure, featuring a hydrophilic shell and a hydrophobic core. The primary structural component is glycol-grafted chitosan, which forms glycolipid-based nanomicelles. A therapeutic agent, selected from hydrophobic drugs and therapeutic peptides, is encapsulated within the hydrophobic core. A targeting ligand, specifically a TGN peptide, is conjugated to the nanocarrier's surface to interact with receptors on the Blood Brain Barrier (BBB), facilitating the delivery of the therapeutic agent to brain tissue. The nanocarrier crosses the BBB, releasing the therapeutic agent sustainably at physiological pH with improved solubility and stability in water The invention includes a method to prepare nanocarriers by forming nanomicelles, encapsulating the therapeutic agent, and conjugating the targeting ligand.
