Sudhakara Rao; P. Manikantha; and Ch. Ramya on August 23, 2026, for Substituted Pyrazolo[3,4-D]pyrimidine Compounds As Nlrp3 Inflammasome Inhibitors And Pharmaceutical Compositions Thereof.

Inventors include Sureshkumar Jaka; Venkata Maruti Prasad S; Maheswara Rao Addepalli; I. Sudhakara Rao; P. Manikantha; and Ch. Ramya.

The application for the patent was published on August 28, 2026, under issue no. 35/2026.

Abstract: The present invention relates to novel substituted pyrazolo[3,4-d]pyrimidine compounds, pharmaceutically acceptable salts, stereoisomers, and pharmaceutical compositions thereof for modulation and inhibition of the NLRP3 inflammasome pathway. The disclosed compounds comprise structurally optimized aryl, heteroaryl, spirocyclic, bridged bicyclic, and fused bicyclic substituents that provide enhanced selectivity, metabolic stability, oral bioavailability, and potential blood-brain barrier penetration. The invention further provides methods for the preparation of the compounds and their therapeutic use in the treatment of inflammatory, autoimmune, metabolic, and neurodegenerative disorders associated with aberrant NLRP3 inflammasome activation, including non- alcoholic steatohepatitis (NASH), Alzheimer’s disease, Parkinson’s disease, gout, and related inflammatory conditions.
