Abstract: Disclosed herein are submicrometre polymeric nanosponges loaded with the antifungal agent eberconazole, and hydrogel compositions incorporating such nanosponges, together with methods for their preparation and evaluation. The nanosponges are prepared from ethyl cellulose and polyvinyl alcohol by an emulsion solvent diffusion process, yielding particles characterised by submicrometre size, favourable zeta potential, low polydispersity and high percentage drug entrapment efficiency, as confirmed by particle size analysis, Fourier transform infrared spectroscopy, differential scanning calorimetry and scanning electron microscopy. The optimised nanosponge formulation is incorporated into a hydrogel base formed from a cross-linked acrylic acid polymer together with penetration enhancing agents, and the resulting hydrogel exhibits acceptable viscosity, pH, swelling behaviour and enhanced in vitro and ex vivo skin permeation relative to a marketed eberconazole formulation. The optimised hydrogel further demonstrates antifungal efficacy comparable to or exceeding that of the marketed product, together with dermal safety confirmed by histological examination. The invention accordingly provides a stable, scalable and industrially applicable topical drug delivery platform for the controlled and sustained treatment of superficial fungal infections, addressing the poor solubility, low bioavailability and hepatic metabolism limitations associated with conventional eberconazole dosage forms
