Abstract: The present invention related to the design of a structure-based method for the discovery of novel marine natural products that act as therapeutic inhibitors of disease- associated proteins. Marine ecosystems contain a large pool of highly diverse yet largely unexplored natural products, which are synthesized by marine algae, seaweeds, sponges, corals, molluscs, marine bacteria, marine fungi, and other marine organisms. The present invention proposes a comprehensive approach for the systematic discovery of marine-derived compounds that possess a potential for acting as inhibitors of disease-associated proteins. A library of marine natural products is generated from marine biological sources and natural product databases, while disease-associated protein targets are selected according to their biological and therapeutic importance. Protein structures are analyzed for the presence of functional or ligand binding sites, while marine-derived compounds are screened via structure-based virtual screening and molecular docking for their binding potential and interaction patterns. Binding energy estimation, molecular interactions, physicochemical properties, drug likeness, pharmacokinetics prediction, and toxicity assessment may then be applied to select therapeutically valuable compounds. Potential candidate compounds can be validated through biochemical, enzymatic, cell-based, or disease-specific biological tests to validate that they inhibit the target proteins in question. This invention can be used in the discovery of inhibitors for cancer, inflammatory, autoimmune, infectious, neurologic, metabolic, cardiovascular, and other diseases associated with the dysregulation of certain proteins. The marine natural compounds discovered in accordance with the invention can be used as therapeutic lead compounds, molecular inhibitors, pharmaceutically active compounds, or scaffold molecules for future development.
