Abstract: The present disclosure relates to steroidal alkaloid compounds identified as candidate dipeptidyl peptidase-4 (DPP-4) inhibitors for the management of type 2 diabetes mellitus (T2DM), and to a computational method for identifying such compounds. The method comprises curating a library of steroidal alkaloid compounds from a chemical structure database, screening the compounds for drug-likeness, performing molecular docking of the compounds against the three-dimensional crystal structure of human DPP-4 enzyme, and further evaluating lead compounds by absorption, distribution, metabolism, excretion and toxicity (ADMET) prediction, metabolic site-of-reactivity prediction, molecular dynamics (MD) simulation, Molecular Mechanics Poisson-Boltzmann Surface Area (MM-PBSA) binding free energy calculation, and density functional theory (DFT) analysis. Using this method, two steroidal alkaloid compounds, Terminamine G (designated SAL1) and Tomatidenol (designated SAL2), were identified as exhibiting predicted binding energies of about -11.2 kcal/mol and -10.8 kcal/mol respectively against DPP-4, more favourable than the standard DPP-4 inhibitor sitagliptin (-9.2 kcal/mol), together with favourable predicted pharmacokinetic, metabolic reactivity and structural stability profiles. The disclosure further provides pharmaceutical compositions comprising these compounds for use in the management of type 2 diabetes mellitus by inhibition of DPP- 4.
