Abstract: The present invention relates to a novel process for extraction, characterization, and pharmacological evaluation of bio-active phytoconstituents obtained from Achyranthes aspera seeds for the treatment and management of asthma. Asthma is a chronic inflammatory respiratory disorder associated with airway hyperresponsiveness, oxidative stress, eosinophilic infiltration, and inflammatory mediator release. The present invention provides a standardized herbal therapeutic approach utilizing hydroalcoholic seed extract of Achyranthes aspera possessing significant anti-asthmatic potential. Seeds procured from Yucca Enterprises, Mumbai were authenticated, shade dried, pulverized, and subjected to hydroalcoholic extraction. The obtained extract was further evaluated through preliminary phytochemical screening, Thin Layer Chromatography (TLC), High Performance Thin Layer Chromatography (HPTLC), DPPH radical scavenging antioxidant assay, and LC-MS analysis for characterization of bio-active phytoconstituents. Identified compounds were further subjected to molecular docking studies against asthma-related molecular targets to validate therapeutic interactions and binding affinity. Pharmacological evaluation was performed using toluene diisocyanate-induced guinea pig models to assess anti-asthmatic activity. Hematological parameters including total WBC count, total leukocyte count, lymphocyte count, eosinophil count, and neutrophil count were analyzed. The extract-treated groups demonstrated significant reduction in inflammatory cell infiltration and oxidative stress markers compared to disease control animals. The novelty of the invention lies in the integrated process combining standardized hydroalcoholic extraction, chromatographic fingerprinting, LC-MS-guided phytochemical characterization, molecular docking validation, antioxidant evaluation, and in-vivo anti-asthmatic assessment of Achyranthes aspera seed phytoconstituents for development of an effective plant-based respiratory therapeutic agent.
