ABSTRACT Title: SN-38 PRODRUGS WITH BUILT-IN NUCLEOPHILE-ASSISTED RELEASE SYSTEMS FOR ENZYME-INDEPENDENT ACTIVATION The present invention relates to SN-38 (D1) prodrugs incorporating Built-In Nucleophile-Assisted Release Systems (BINARS) linkers for enzyme-independent release of SN-38. Seventy-three compounds (A-1 to A-73) are disclosed across three structural classes under Formula (I): Type 1 disulfide-containing BINARS–SN-38 compounds for thiol-triggered release, Type 2 BINARS–SN-38 compounds for pH-dependent intramolecular cyclization, and Type 3 BINARS–SN-38 compounds containing PABC self-immolative spacers for antibody-drug conjugate applications. Lead compound A- 42 provides rapid SN-38 release under physiological conditions, and lead compound A-59 provides improved stability for ADC payload applications. The invention further provides parallel platform extension configurations under Formula (VI) covering alternative camptothecin analogues (D2–D7). The invention further provides CL3EA linker-drug conjugates, antibody-drug conjugates, Type 4 dicarboxylic acid-Val-Cit-PABC-Type 2 BINARS–SN-38 conjugates, pharmaceutical compositions, and the use of these compounds, conjugates, or compositions in the treatment of cancer, including personalized therapy utilizing Type 2 BINARS– SN-38 compounds stratified proactively based on UGT1A1 genotyping.
