Abstract: The present invention is a validated analytical method for the simultaneous estimation of gallic acid, quercetin and diosgenin, three chemically diverse markers of divergent polarity, in successive n-hexane, ethyl acetate and ethanolic extracts of the leaves, fruit and stem of Pedalium murex. In one aspect, an HPTLC method employs a mobile phase of toluene:ethyl acetate:formic acid:methanol (4:4:1:1, v/v/v/v), with gallic acid and quercetin scanned at about 270 nm and diosgenin scanned at about 370 nm following anisaldehyde—sulphuric acid derivatization, resolving the markers at Rf values of about 0.58, 0.74 and 0.90 respectively. In another aspect, a gradient RP-HPLC method on a C8 column, using a water: acetonitrile mobile phase programmed from about 45% to about 95% acetonitrile at 1 mL/min with detection at about 205 nm, resolves the three markers with retention times of about 1.15, 3.05 and 11.41 minutes respectively within about 13 minutes. Both methods are validated as per ICH Q2 (R2) guidelines for linearity, sensitivity, precision and robustness, and provide a rapid, reliable basis for the standardization and quality control of Pedalium murex raw material, extracts.
