Abstract: The present invention relates to sesamol-amine hybrid compounds of Formula (I), and pharmaceutically acceptable salts, solvates, hydrates and stereoisomers thereof, together with a process for their preparation. The compounds comprise a sesamol-derived benzo[d][1,3]dioxol-5-yloxy moiety linked through a 2-hydroxypropoxy chain to a nitrogen-containing moiety. The process comprises nucleophilic ring opening of an oxirane-substituted sesamol intermediate with selected secondary amines to afford the corresponding sesamol-amine hybrid compounds. Representative compounds comprise pyrrolidine, piperidine, piperazine, benzylpiperidine, benzylpiperazine, and related amino substituents. The disclosed compounds exhibited acetylcholinesterase inhibitory activity, supported by molecular docking, density functional theory, and molecular dynamics simulation studies. Among the disclosed compounds, SES-1BP, SES-4BP and SES-NBA demonstrated particularly promising acetylcholinesterase inhibitory activity. The invention provides sesamol-amine hybrid compounds and a process for their preparation useful for the development of acetylcholinesterase-targeted pharmaceutical agents. FIG. 1
