Abstract: ABSTRACT SERUM-FREE CELL CULTURE MEDIA COMPOSITION FOR CHINESE HAMSTER OVARY CELLS AND PREPARATION METHOD THEREOF The present invention provides a serum-free, chemically defined cell culture medium for Chinese Hamster Ovary (CHO) cells, comprising amino acids, vitamins, inorganic salts, trace elements, carbohydrates, growth-enhancing additives, bioactive additives, and media conditioning agents, all of non-animal origin. The invention provides a temperature-controlled manufacturing method comprising: selecting heat-stable components for initial processing; blending in a cooling jacket- equipped blender to form a premix; transferring to a pin mill with liquid nitrogen cooling; milling with temperature monitoring then cooling; adding thermolabile components to the cooled material; transferring to a low-shear mixer with cooling; blending with real-time temperature logging; and sampling from multiple zones to verify blend uniformity. The disclosed medium supports high-density cell growth, enhanced protein expression, and demonstrates excellent batch-to-batch reproducibility. The invention addresses prior art limitations regarding thermolabile component preservation, batch consistency, and contamination risks, providing a cost-effective alternative for biosimilar and monoclonal antibody manufacturing.
