Abstract: ABSTRACT The present invention provides a green, sequential, single-vessel process for extraction of anthocyanin pigments from plant biomass or agricultural waste biomass. The process comprises enzymatically pretreating the biomass with cellulase under conditions suitable for enzymatic cell-wall disruption, followed by direct addition of a Natural Deep Eutectic Solvent (NADES) into the same vessel and ultrasound-assisted extraction. The process intentionally exploits the intrinsic acidity and relatively low pH of the NADES to suppress cellulase activity, thereby providing in-situ enzyme deactivation without requiring an intermediate enzyme separation, washing, or deactivation step. Ultrasound-induced acoustic cavitation further enhances cell disruption, solvent penetration, mass transfer, and anthocyanin release. The sequential integration of cellulase pretreatment, NADES extraction, and ultrasound treatment improves extraction efficiency while reducing solvent consumption, processing complexity, and environmental impact. The process is performed under mild temperature conditions to minimize thermal degradation and preserve anthocyanin stability, providing extracts suitable for food colorants, nutraceuticals, functional products, and bioactive packaging applications.
