Abstract: The present invention relates to a self-calibrating smart transdermal patch for adaptive and controlled delivery of a therapeutic agent. The patch comprises a drug- containing reservoir or matrix, at least one sensor, a controllable drug-release actuator, a processing unit, a power source, and an integrated feedback control system. Upon application, the patch establishes baseline sensor values and generates one or more calibration parameters. During use, sensor measurements associated with the skin interface, patch condition, physiological status, and/or drug-release performance are compared with stored baseline values and predetermined targets. The processing unit automatically updates calibration parameters to compensate for sensor drift and changing operating conditions and regulates the drug-release actuator to increase, decrease, maintain, pause, or terminate delivery. The actuator may include an iontophoretic system, micropump, microvalve, electrically responsive membrane, thermal element, microneedle-assisted system, or combinations thereof. Safety limits, fault detection, data logging, and optional wireless communication may also be provided. The invention thereby provides a closed-loop transdermal drug-delivery platform capable of improving delivery consistency, adaptability, and operational safety.
