Abstract: The present invention relates to the field of medicinal chemistry, specifically to the development of selective SGLT-2 inhibitors for managing glycemic levels in Type-2 Diabetes. The invention comprises a core structure based on benzothiazole or benzofuran scaffolds, integrated with a bioisosteric analog selected from thiadiazole or oxadiazole rings. Additionally, one or more hydrophobic moieties, chosen from aromatic or alkyl groups, are attached to the core structure. These components are configured to selectively inhibit SGLT-2 over SGLT-1, enhancing the specificity and efficacy of the inhibitor. The invention also includes a method for preparing these inhibitors, involving molecular docking studies to predict binding modes and interactions with the SGLT-2 active site, ensuring high selectivity and effectiveness in glycemic control.
