The application for the patent was published on August 28, 2026, under issue no. 35/2026.

Abstract: The present invention relates to a saliva-based competitive enzyme-linked immunosorbent assay (ELISA) platform for quantitative determination of progesterone in human saliva. The method provides a non-invasive alternative to conventional blood-based progesterone measurement. A saliva sample is collected and processed using a progesterone-OVA coated ELISA plate blocked with approximately 1% BSA. Mouse anti-progesterone primary antibody is introduced according to a competitive assay format, followed by goat anti-mouse enzyme-labelled secondary antibody. Washing is performed between assay stages to remove unbound components and reduce non-specific binding. Following substrate addition, the developed colour is measured spectrophotometrically using a microplate reader. Progesterone concentration is determined by comparison with a standard curve generated using known progesterone standards. The method enables repeated progesterone monitoring and is applicable to menstrual cycle documentation, fertility assessment, ovulation monitoring, luteal phase assessment, reproductive endocrinology and related reproductive health applications. The platform may further be adapted for diagnostic ELISA kits and future portable or point-of-care systems.
