Abstract: The present invention is a pharmaceutical cocrystal comprising rifampicin and quercetin in an approximately one-to-one molar relationship, prepared from 1.6 g rifampicin and 0.6 g quercetin by ethanol-assisted solvent evaporation. The constituents form a new non-covalently stabilized crystalline phase distinguished from the individual materials and their physical mixture by shifted hydroxyl and carbonyl bands, altered powder X-ray diffraction peaks, and compact plate-like morphology. The cocrystal provides faster rifampicin dissolution in acidic and phosphate-buffer media and lower degradation in simulated gastric fluid with and without pepsin. In a rifampicin-induced hepatotoxicity study, the cocrystal reduced total bilirubin, AST, ALT, and ALP values toward normal levels and preserved body weight during oral administration. A preparation process uses controlled stirring, evaporation, solvent recovery, temperature sensing, mass-loss monitoring, and vapor sensing to obtain reproducible batches. The invention therefore provides a physical pharmaceutical solid form combining gastric protection, improved drug release, and quercetin-derived hepatoprotection without covalently modifying rifampicin.
