Abstract: The present invention relates to the development of an assay for the identification of species and sex of the cattle in tissue/ meat or related samples. It particularly relates to design and select the primers for conventional and real time PCR assays for the identification of species and sex of the cattle accurately in tissue/ meat or related samples. It specifically relates to the development of a rapid, accurate and sensitive assay for the identification of species and sex of the cattle in tissue/ meat or related samples by combining the conventional PCR, SYBR Green–based qPCR and probe-based qPCR in a sequential and confirmatory manner. The invention also relates to conventional PCR and real time PCR assays targeting mitochondrial D loop for the identification of cattle species and targeting SRY and AMELX gene for the determination of the sex from meat samples by performing sex determination after the species authentication. The invention further relates to the kit and method to perform the identification of species and sex of the cattle in tissue/ meat or related samples.
