Abstract: ABSTRACT Pyridine and Pyrimidine heterocycles as Piperazine-Chalcone Hybrids The present invention relates to a novel class of nitropyridine and methoxy pyrimidine with piperazine-chalcone hybrid compounds represented by Formula-I (A-J), wherein a Pyridine and pyrimidine moiety is linked to a piperazine ring and further connected to a chalcone framework bearing substituted phenyl groups selected from 4-chloro, 4-fluoro, 4-nitro, 3-chloro, 3-fluro, 2-chloro, and 4-cyanophenyl substituents. The invention further relates to an efficient synthetic process for the preparation of said compounds through a microwave-assisted C-N bond-forming reaction between 2-chloro-5-nitropyridine and 2-chloro-5-methoxypyridine with 1-(4- (piperazin-1-yl)phenyl)ethan-1-one in the presence of 1-butyl-3- methylimidazolium acetate ([BMIM]OAc) and dimethylformamide (DMF) and DIPEA and PEG-400 respectively, followed by Claisen-Schmidt condensation of the resulting intermediate with substituted aromatic aldehydes under alkaline conditions. The invention also encompasses pharmaceutical compositions comprising one or more of the disclosed compounds together with pharmaceutically acceptable carriers, excipients, or diluents. The synthesized compounds may exhibit biological activities including anticancer, antimicrobial, anti-inflammatory, antioxidant, antiviral, and enzyme inhibitory properties, thereby providing promising candidates for the development of nextgeneration therapeutic agents.
