Abstract: The present invention provides a one-pot multicomponent process for the preparation of Mannich base derivatives of diethyl malonate. An aromatic aldehyde, such as anisaldehyde or vanillin, and an aromatic amine, such as aniline or 3-chloroaniline, are stirred in a lower alcohol solvent, namely methanol or ethanol, with a catalytic amount of a mineral acid at room temperature; diethyl malonate, ammonium chloride and tetrabutylammonium bromide (TBAB) are then added, and the mixture is stirred at room temperature for about 20 to 25 hours. The product is isolated by aqueous work-up, solvent extraction and column chromatography. In contrast to the solid Mannich bases of the prior art, the process consistently affords the derivatives as stable oily compounds in good yield (about 75–76%) and purity under mild conditions using inexpensive catalysts. The invention further provides the compounds so obtained, including diethyl 2-((4-hydroxy-3- methoxyphenyl)(phenylamino)methyl)malonate, and their use as antibacterial agents and synthetic intermediates.
