Abstract: ABSTRACT The present invention relates to novel fluoride-free dual-action paediatric toothpaste formulation, designated Probiodent, comprising Lactobacillus salivarius at 1.0% w/w and nano-hydroxyapatite at 5.0% w/w as synergistic active ingredients in a stable pharmaceutical paste base of calcium carbonate, sorbitol, precipitated silica, cellulose gum, xanthan gum, sodium lauryl sulphate, sodium silicate, sodium saccharin, benzyl alcohol, and mint-clove flavouring, prepared by sterile trituration. The formulation exhibits a pH of 7.8 to 8.1, viscosity of 42,000 to 47,000 cps, spreadability of 6.1 to 6.4 g.cm2/sec, and foaming ability of 43 to 48 ml. The probiotic active agent demonstrates significant antibacterial activity against Streptococcus mutans with a mean zone of inhibition of 19 mm. Scanning electron microscopy with energy dispersive spectroscopy confirms enamel remineralization equivalent to fluoride-based toothpaste. Animal safety studies confirm non-irritancy. The invention addresses the critical pediatrics oral health need for a fluoride-free caries-preventive product safe for children under twelve years of age.
