Abstract: PRIMERS AND GUIDE RNA SEQUENCE FOR THE DETECTION OF HUMAN PAPILLOMAVIRUS TYPE 18 (HPV 18) BY RPA-CRISPR-CAS12A BASED FLUORESCENT ASSAY Cervical cancer is a serious threat worldwide, as it is the fourth most common cancer among women. It is mainly caused by the Human papillomavirus (HPV). HPV 18 is one of the high-risk types that is responsible for cervical carcinoma. Traditional screening techniques have limited sensitivity, and thus, molecular detection of HPV has been introduced to for early detection and to reduce the mortality rate. In our study, recombinase polymerase amplification (RPA) integrated with a CRISPR-Cas12a assay with fluorescence endpoint detection, has been developed. The fluorescence results can be visualized under UV light. This fluorescence endpoint detection system is highly sensitive, detecting up to ~4 copies/µL of DNA. Unlike conventional methods such as polymerase chain reaction (PCR), the developed fluorescence detection method takes approximately an hour. Thus, the developed RPA-CRISPR-Cas12a assay offers a promising, reliable, and rapid solution for mass screening for HPV 18 detection, even in low-resource settings.
