Abstract: Skin diseases including psoriasis, dermatitis, eczema, and infectious skin conditions impose substantial global health burden. Berberine, an isoquinoline alkaloid with anti-inflammatory, antimicrobial, antifungal, and antioxidant properties, has limited topical applicability due to poor skin permeability, low aqueous solubility, and rapid degradation. The present invention discloses a topical pharmaceutical formulation comprising berberine loaded into chitosan-coated poly(lactic-co-glycolic acid) nanoparticles dispersed in a carbopol hydrogel matrix. The optimized nanoparticles exhibited mean particle size of 187 nanometers, encapsulation efficiency of 78.6 percent, and positive zeta potential of 28.3 millivolts. The formulation demonstrated sustained drug release over 24 hours and 3.82-fold enhanced dermal deposition compared to conventional gel. In vivo evaluation in an imiquimod-induced psoriasis mouse model showed significant reduction in disease severity scores, epidermal hyperplasia, and inflammatory cytokines comparable to betamethasone without skin atrophy, establishing a safe and effective nanoformulation-based alternative for dermatological therapy.
