Abstract: ABSTRACT The present invention relates to a predictive and experimentally validated pharmaceutical formulation optimization system for a selective vincristine derivative. The invention integrates molecular docking affinity with pharmaceutical formulation variables and drug-release kinetic parameters to identify an optimized controlled-release formulation. The selective vincristine derivative was subjected to molecular docking against a predetermined therapeutic target, preferably tubulin, and demonstrated favorable predicted molecular interaction. The derivative was incorporated into a biodegradable nanocarrier and a plurality of formulations were prepared by varying carrier and stabilizer concentrations. The formulations were characterized for particle size, polydispersity, zeta potential, encapsulation efficiency, drug loading, morphology and in-vitro drug release. Release profiles were fitted to zero-order, first-order, Higuchi, Korsmeyer-Peppas and Weibull models. A predictive optimization model incorporating docking affinity and release kinetic parameters was generated to identify an optimized formulation. The predicted formulation was experimentally prepared and demonstrated close agreement between predicted and observed particle size, encapsulation efficiency and drug-release characteristics. The invention therefore provides a computationally guided and experimentally validated platform for optimization of controlled-release pharmaceutical formulations of selective vincristine derivatives.
