Abstract: The present invention relates to a polymeric transdermal drug delivery system incorporating Remogliflozin Etabonate for sustained treatment of diabetes mellitus. The transdermal patch comprises hydroxypropyl methylcellulose, Eudragit RLPO, Eudragit RSPO, a polyvinyl alcohol backing membrane, and a suitable plasticizer to regulate drug release and maintain mechanical stability. The patch is fabricated using a solvent casting and evaporation technique to produce a flexible film for prolonged skin application. Formulation variables are optimized through a Box–Behnken experimental design to achieve desirable physicochemical properties and sustained drug release. The developed system enables controlled transdermal permeation of Remogliflozin Etabonate over an extended period. In vivo studies in streptozotocin-induced diabetic animals demonstrate improved glycemic control and biochemical outcomes. The invention provides a reproducible, sustained-release transdermal therapeutic platform for long-term antidiabetic therapy.
