Abstract: POLYMERIC NANOPARTICLE COMPOSITIONS FOR ENCAPSULATION AND SUSTAINED RELEASE OF HYDROXYZINE HYDROCHLORIDE The present invention relates to polymeric nanoparticle compositions for encapsulation and sustained release of Hydroxyzine Hydrochloride. The invention provides biodegradable and biocompatible polymeric nanoparticles comprising polycaprolactone and polyhydroxybutyrate for controlled drug delivery. The nanoparticles are prepared using a solvent evaporation method and optimized using a Box-Behnken Design. The formulation exhibits an initial burst release followed by sustained drug release over approximately 24 hours, thereby reducing the frequent administration associated with conventional Hydroxyzine Hydrochloride tablets, capsules, and oral solutions. The nanoparticle compositions improve therapeutic efficacy, reduce dosage frequency, and enhance patient compliance. The invention further provides a simple, scalable, and economical manufacturing process suitable for pharmaceutical applications.
