Abstract: ABSTRACT TITLE: PHYTOCHEMICAL AND PHARMACOLOGICAL EVALUATION OF ETHANOLIC LEAF EXTRACT OF VITIS VINIFERA FOR NOOTROPIC ACTIVITY The present invention relates to the phytochemical and pharmacological evaluation of an ethanolic leaf extract of Vitis vinifera L. (ELEVV) for nootropic activity. The extract, prepared through Soxhlet extraction with ethanol following petroleum ether defatting, exhibits a rich phytochemical profile comprising phenolics, flavonoids, tannins, saponins, and alkaloids, with total phenolic content of 413.88 mg GAE/g and total flavonoid content of 156.68 mg QE/g. ELEVV demonstrates potent antioxidant activity with a DPPH IC50 of 40.50 µg/mL. Acute oral toxicity studies confirm safety at 2000 mg/kg. In scopolamine-induced amnesia in Wistar rats, chronic administration of ELEVV at 100, 200, and 400 mg/kg for 21 days significantly improves spatial learning and memory in the Morris Water Maze test dose-dependently, with 400 mg/kg approaching donepezil efficacy. The extract concurrently inhibits brain acetylcholinesterase activity, preserving cholinergic neurotransmission. These findings support Vitis vinifera leaf extract as a promising natural nootropic candidate for Alzheimer’s disease management.
