Abstract: The present invention relates to a physically authenticated DNA data storage system with Nano-Puf-based tamper-evident security for secure, high-density, and long- term storage of digital data. Digital information is encoded into DNA oligonucleotide sequences with error correction and encapsulated within a protective medium for long-term preservation. Each storage capsule incorporates embedded nanoparticles that generate a unique nano-scale physically unclonable function (Nano-PUF), providing a distinct physical identity for authentication. The system verifies the authenticity of the storage capsule using a Nano-PUF-based authentication process to prevent unauthorized access, tampering, cloning, or substitution. The Nano-PUF signature is cryptographically associated with the stored data to ensure integrity and traceability. Upon successful authentication, the DNA is sequenced and decoded to reconstruct the original digital information. Accordingly, the invention provides a secure, tamper-evident, and physically authenticated DNA-based archival platform for long-term digital data preservation
