Abstract: The present invention discloses a peppermint essential oil based nanoemulsion composition and a method for its preparation and application against multidrug resistant Ureaplasma spp. The nanoemulsion comprises peppermint essential oil, Tween 80 as surfactant, ethanol as co-solvent, and distilled water, prepared by spontaneous emulsification combined with ultrasonication. The formulation exhibits a mean particle size of 212.4 nm, a zeta potential of minus 32.4 mV, and retains the functional groups of the essential oil. The nanoemulsion demonstrates significant antibacterial activity against MDR Ureaplasma spp. clinical isolates with MIC values of 41.66 to 83.33 micrograms per millilitre, comparable to standard antibiotics. It also exhibits concentration-dependent biofilm inhibition up to 68.28 percent at MIC and anti- quorum sensing activity with a 25 mm zone of inhibition against Chromobacterium violaceum. The invention provides an alternative therapeutic strategy for managing urogenital infections caused by multidrug resistant Ureaplasma spp.
