Abstract: The present invention relates to sustained-release oral tablets of paracetamol using in-situ polyelectrolyte complex (PEC) formation. The tablet comprises paracetamol and a PEC-forming polymer pair including chitosan (cationic) and sodium starch glycolate (anionic). A chitosan binder solution is prepared as about 1% w/v in about 1% acetic acid, cooled to about 4°C and neutralized using sodium bicarbonate, and used for wet granulation of paracetamol with sodium starch glycolate and excipients, followed by sieving, drying, lubrication and compression. On exposure to acidic medium (about pH 1.5), chitosan and sodium starch glycolate form a dense PEC gel barrier that limits burst release and sustains drug diffusion. Dissolution testing using staged media (pH 1.5 for 2 h, pH 6.8 for 8 h, pH 7.5 for 2 h) demonstrates sustained release up to about 12 hours. The tablets improve dosing convenience and patient compliance. (Accompanied Figure No. 1)
