Abstract: The present invention relates to optimized polymeric nanoparticles of Baricitinib prepared using nanoprecipitation method and optimized through Box-Behnken design. The formulation variables include polymer amount, surfactant concentration, and stirring speed. The polymeric nanoparticle formulation, comprising: Baricitinib as an active pharmaceutical ingredient; Ethyl cellulose as a polymer in an amount of about 250 mg; a surfactant comprising Tween 20 in a concentration of about 2%; and stirring speed of about 2400 rpm; wherein the nanoparticles having an average particle size of about 126.4 nm. The nanoparticles exhibit an entrapment efficiency of about 98.343 ± 0.46%, a percentage yield of about 92.624 ± 0.43%, in-vitro drug release of about 60.818 ± 0.28% over 8 hours, a zeta potential of about -14 mV and a polydispersity index of about 0.444. The method for optimizing a Baricitinib nanoparticle formulation, comprising: selecting formulation variables including: amount of ethyl cellulose, surfactant concentration, and stirring speed; applying a Box-Behnken design with three levels (-1, 0, +1); evaluating responses comprising: percentage yield, entrapment efficiency, and in-vitro drug release; identifying an optimized formulation based on maximum yield and entrapment efficiency and minimum drug release. The optimized nanoparticles exhibit improved entrapment efficiency, controlled drug release, and stability, making them suitable for pharmaceutical applications.
