Abstract: Optimized Gastroretentive Floating Tablet Formulation of Pantoprazole Sodium for GERD Treatment This invention describes a gastroretentive floating tablet formulation of Pantoprazole Sodium for the effective treatment of gastroesophageal reflux disease (GERD) and related gastric disorders. The formulation incorporates hydrophilic polymers, such as HPMC K100M and HPMC K15M, to form a gel matrix that ensures sustained drug release and prolonged gastric retention. Sodium bicarbonate serves as a gas-generating agent, creating buoyancy by producing carbon dioxide upon reaction with gastric acid, allowing the tablet to remain afloat in the stomach for over 9 hours. The tablet demonstrates a floating lag time of less than 30 seconds and a drug release profile of 67.09% within 6 hours, meeting USP standards for controlled drug delivery. The formulation is manufactured using direct compression for scalability and reproducibility. This invention enhances bioavailability, reduces dosing frequency, and improves patient compliance, offering a superior therapeutic solution for GERD management.
