The application for the patent was published on October 09, 2026, under issue no. 41/2026.

Abstract: The present invention relates to an anti-hypertensive active pharmaceutical ingredient (API), Olmesartan Medoxomil. The present invention particularly relates to a co- crystal of olmesartan medoxomil with nicotinamide as a co-former in a ratio of 1:1, to enhance solubility, bioavailability and efficacy of olmesartan medoxomil. The resulting co-crystal exhibited distinct DSC and PXRD profiles and a hydrogen-bonded network in the triclinic P1 system. Compared to olmesartan medoxomil, the co- crystal showed a 7.0-fold increase in solubility, 80% drug release within 24 hours, and a 6.6-fold enhancement in relative bioavailability. In DOCA-salt-induced hypertensive rats, it significantly reduced (p 0.05) blood pressure and heart rate within one week. Additionally, improved antioxidant and anti-inflammatory activity was evidenced by significantly increased (p 0.01) glutathione, superoxide dismutase, and nitrite levels, along with reduced lipid peroxidation and pro-inflammatory cytokines.
