Abstract: The present disclosure provides a nucleotide sequence encoding RPGRORF15. Said nucleotide sequence is codon optimized for higher stability, fidelity in replication and higher expression levels compared to the wild-type sequence. The present disclosure further provides a nucleotide construct comprising the codon optimized nucleotide sequence encoding RPGR0RF15 along with 10 various regulatory elements such as promoter, one or more UTR(s) and poly A signal. Further provided in the present disclosure is a recombinant vector comprising the nucleotide construct and host cell comprising the recombinant vector. The present disclosure also envisages a composition for treatment or prophylaxis of Retinitis Pigmentosa, particularly, XLRP and method(s) for the same.
