Abstract: This invention relates to a novel synergistic topical pharmaceutical composition of Tapinarof formulated with functional excipients containing a combination of 10 polyethers (PEG 400 and PEG 4000), PLGA, and 4-chlororesorcinol for the treatment of psoriasis. Tapinarof acts by modulating aryl hydrocarbon receptors and reducing pro-inflammatory cytokine production. The selected excipients enhance drug penetration, stability, and therapeutic efficacy. Polyethylene glycols provide humectant, anti-inflammatory, emollient, and penetration-enhancing properties, 15 while PLGA minimizes irritation and burning, offering a soothing, sustained-release effect. 4-Chlororesorcinol confers antioxidant and photoprotective benefits, preventing UV-induced skin damage. As evident from animal study synergistic formulation showed enhanced keratolytic and anti-inflammatory activity, promoting better resolution of hyperkeratotic psoriatic lesions. This non-steroidal, 20 anti-inflammatory, and UV-protective formulation provides a more effective therapeutic alternative for the management of chronic psoriatic inflammation.
