Abstract: ABSTRACT The present invention relates to novel quinoline-based bis-oxadiazole derivatives, their process of preparation, and their application as anticancer agents. The compound comprises a quinoline ring linked to a first oxadiazole ring, the first oxadiazole ring being further linked, through a sulfanylmethylene bridging group, to a second oxadiazole ring bearing a substituted phenyl ring. The compound is prepared by a multi-step process comprising esterification, hydrazide formation, cyclization, alkylation, amidoxime formation, and coupling with a substituted aromatic acid followed by cyclization. The compounds demonstrate significant in vitro cytotoxic activity against breast and colon cancer cell lines, and exhibit favourable binding interaction with the epidermal growth factor receptor tyrosine kinase domain, as confirmed by molecular docking studies, together with favourable pharmacokinetic properties. It provides a pharmaceutical composition comprising the said compound and a method of treating cancer by administration thereof, thereby providing potential candidates for pharmaceutical applications.
