Abstract: ABSTRACT NOVEL PYRROLIZINE-BASED ACETAMIDE COMPOUNDS AND SYNTHESIS THEREOF The present invention relates to a novel pyrrolizine- based acetamide compound, 2-(6-(4-chlorophenyl)-2,2-dimethyl-7-phenyl-2,3-dihydro-1H-pyrrolizin-5-yl)-N-(4-nitrophenyl)acetamide (I), derived from licofelone by carbodiimide-mediated coupling with 4-nitroaniline. The compound demonstrates potent in vitro cytotoxicity against gastric (GSU) and pancreatic (PANC-1) cancer cell lines, with while exhibiting reduced cytotoxicity towards normal cells. Molecular docking studies confirm favourable binding of the compound to Aurora Kinase A (AURKA1), supporting its potential mechanism of anticancer action.
