Abstract: The present invention relates to novel pyrazole derivatives exhibiting potent anti-arthritic activity. Rheumatoid arthritis (RA) is a chronic autoimmune disorder causing joint inflammation, cartilage degradation, and disability. Among synthesized derivatives, 3,4-bis(3-chlorophenyl)-5-methyl-1H-pyrazole was identified as a lead compound based on in-silico molecular docking and molecular dynamics studies targeting TNF-a. This compound was further evaluated in vivo using complete Freund’s adjuvant (CFA)-induced arthritis in Wistar rats. Treatment significantly reduced paw swelling, paw volume, and arthritic index while improving body weight, mobility, and gait. Biochemical analyses demonstrated reduced oxidative stress markers, including glutathione, catalase, superoxide dismutase, and lipid peroxidase, along with downregulation of proinflammatory cytokines TNF-a and IL-6. Histopathological and radiological studies confirmed decreased synovial hyperplasia, inflammatory infiltration, and joint erosion. The compound displayed favorable pharmacokinetic and ADMET profiles, indicating potential as a safe, effective anti- arthritic agent. This invention provides a novel therapeutic approach for RA with enhanced efficacy and improved safety compared to conventional NSAIDs.
