Abstract: Novel Process of Production of L-Asparaginase The present invention relates to a novel process for production of extracellular L-asparaginase using Brevibacillus borstelensis ML12, isolated from soil and identified through morphological, biochemical, physiological and 16S rDNA characterization. The process comprises screening soil-derived bacterial isolates for L-asparaginase production, selecting the identified strain, and cultivating the strain in an L-asparagine-containing fermentation medium under optimized conditions of pH, temperature, culture age, medium volume, fermentation time and agitation. The extracellular enzyme is recovered from the fermented broth and purified by ammonium sulphate precipitation followed by dialysis. The purified L-asparaginase exhibits favourable enzymatic and kinetic characteristics, including a specific activity of approximately 523.8 IU/mg, purification fold of 1.94 and Km of approximately 0.253 mM. The produced enzyme further demonstrates anticancer activity against triple-negative breast cancer cells, particularly MDA-MB-231 cells, with investigation of apoptosis and intracellular reactive oxygen species generation.
