The application for the patent was published on September 11, 2026, under issue no. 37/2026.
Abstract: The present invention relates to novel imipridone derivatives for the treatment of aggressive cancers, particularly triple-negative breast cancer (TNBC). The derivatives are structurally optimized to target mitochondrial caseinolytic protease P (HsClpP) and are synthesized through a two-step process involving formation of substituted cyclic guanidine intermediates followed by cyclocondensation with a piperidone derivative. Compounds S1–S5 are characterized using FTIR, ¹H NMR, ¹³C NMR, LC- MS, chromatographic purity analysis, and melting-point determination. Representative compounds S2 and S4 exhibit high purity and promising cytotoxic and antiproliferative activity against human breast cancer cells. Biological evaluation demonstrates induction of G2/M-phase cell-cycle arrest, apoptosis, mitochondrial membrane depolarization, and associated mitochondrial dysfunction. The compounds thereby inhibit cancer-cell proliferation and promote programmed cell death through complementary mechanisms. The invention provides synthetically accessible, structurally optimizable, and biologically validated imipridone derivatives, particularly S2 and S4, as promising therapeutic candidates for further preclinical and pharmaceutical development against TNBC and other aggressive malignancies.
