Abstract: The present invention relates to a novel oil-in-water herbal nanoemulsion pharmaceutical composition for targeted anti-inflammatory therapy comprising curcumin at 0.5 to 3% w/w, Boswellia serrata extract standardized to 65% boswellic acids at 0.5 to 2% w/w, medium-chain triglyceride oil phase, Tween 80 and Span 80 as surfactants, and a folate-polyethylene glycol-distearoyl phosphoethanolamine conjugate as targeting ligand. The nanoemulsion exhibits globule size of 50 to 95 nanometers, polydispersity index below 0.25, encapsulation efficiency above 95%, and selectively targets folate receptor-beta on activated macrophages at inflammatory sites. In preclinical studies, the composition demonstrated 4.3-fold enhanced macrophage uptake, 82% reduction in tumor necrosis factor-alpha, and 71.8% inhibition of carrageenan-induced paw edema in rats, exceeding standard diclofenac. The formulation maintains stability over 12 months and offers a safe, efficacious therapeutic option for rheumatoid arthritis, inflammatory bowel disease, and psoriasis.
