Abstract: ABSTRACT The present invention provides a pharmaceutical composition in the form of abilayer tablet, comprising: an immediate release layer comprising of 50 mg ofesomeprazole; and Extended-release layer comprising of 250 mg Tranexamic acidand pharmaceutically acceptable excipients wherein the pharmaceuticallyacceptable excipients are Calcium carbonate: Citric acid;HPMC; Gum acacia; Microcrystalline cellulose; Sodium starch glycolate; Ethyl cellulose; Carboxymethyl cellulose; Sodium alginate; Eudragit. Bi layer tablet thickness ranged from 4.l5±0.05mm to 5.02±0.0lmm. Hardness ranged from 4.2±0.083kg/cm2to 5.8±0.125kg/cm2. Friability ranged between 0.420 ± 0.002% and 0.639±0.002%. Acid neutralization of esomeprazole layer was excellent in four formulations ranging between 3.20mEq and 5.40mEq. The disintegration time ranged between l2.52±0.62 and l7.82±0.89 minutes. Oral administration of this pharmaceutical composition is preferred. This bilayer tablet containing proton-pump inhibitor and anti-fibrinolytic is highly effectively in treating gastric bleeding responsively.
